چکیده

Hypersensitivity to drugs may lead to the development of drug-induced platelet antibodies and consequently thrombocytopenia and bleeding, usually in the form of purpura. The association of drug and purpura was reported by Vipan(1) even before the role of platelets in haemostasis was recognised. The relationship between drug ingestion and thrombocytopenia was demonstrated by Rosenthal in 1928 when he rechallenged a patient who had just recovered from quinine-induced thrombocytopenia

Drug-antibody-platelet interaction in quinine- and quinidine-induced thrombocytopenia.

Binding of quinine- and quinidine-dependent antibodies to platelets was studied using an electroimmunoassay to measure platelet-bound IgG. Antibodies from four patients with drug-induced thrombocytopenia differed significantly in their interaction with platelets: association constants for binding to platelets at high drug concentrations ranged from 0.29 to 2.6 x 10(7) M(-1), the maximum number ...

Platelet antigens and antibodies in multitransfused patients

Abstract Background and Objectives Following incompatible blood transfusions, anti-HLA and  anti- HPA antibodies may develop and cause various disorders such as post-transfusion purpura, platelet refractoriness, and thrombocytopenia leading to bleeding. The aim of this study was to investigate platelet antigens and antibodies in multi-transfused patients.   Materials and Methods In this desc...

Evaluation of platelet antigens and antibodies in patients with platelet refractoriness

Abstract Background and Objectives Immune platelet refractoriness is a complication of blood transfusion in patients with multiple platelet transfusions. Antibodies against surface antigens, including human platelet antigens (HPAs) and human leukocyte antigens (HLAs), can be produced which can cause the degradation of transfused platelets by macrophages. In this study, the frequency of platele...

Structural basis for quinine-dependent antibody binding to platelet integrin aIIbb3

• Quinine binds to quininedependent antibodies first and causes CDR loop reconfiguration. • A hybrid paratope consisting of quinine and reconfigured CDR recognizes its target epitope. Drug-induced immune thrombocytopenia (DITP) is caused by antibodies that react with specific platelet-membrane glycoproteinswhen the provokingdrug is present.More than 100 drugs have been implicated as triggers fo...

AET-treated platelets: their usefulness for platelet antibody detection and an examination of their altered sensitivity to immune lysis.

2-Aminoethylisothiouronium bromide (AET) increases the sensitivity of blood cells to complement-mediated immune lysis. We compared the sensitivities of untreated or AET-treated platelets to immune lysis induced by different types of platelet antibody in the 51Cr platelet lysis test. AET platelets were 8-16 times more sensitive to autoantibody and alloantibody, but 8-16 times less sensitive to d...
